Best bullet choice, what rate of twist, what powder and how much?

I make my dies and chamber my barrels Im just looking for some intel on the cartridge choice.
(One of Reddings type S 6BR dies could probly be opened up easy enough and hand seat dies get cheap when you have the chamber reamer).
